Benefits: Reliability (emissions controls are nothing but a liability for the owner; stock firmware code mandated by the EPA causes the vehicle to refuse to start if the emissions systems aren't functioning), less hassle, better engine longevity thanks to the required PCM tune that also disables EGR, and mild improvements to fuel economy.

Drawbacks: if you live in an emissions inspection state it might be a bit difficult to pass (haha), cost, and you can kiss your warranty goodbye.

Oh, and I guess you can "roll coal" with a full delete. That can be a benefit or drawback depending on your perspective.

The GDE Hot Tune is an intermediate step between stock and a full delete. It disables some of the emissions systems via code and tweaks the operation of the emissions systems that are still active.

Another thing I'd like to elaborate on, aside from what roadkill said, is that without a DPF you no longer have need of a regen cycle. That not only saves you a little more fuel but it also helps prevent fuel /oil dilution. The regens work by injecting fuel into the exhaust stroke and this can result in contaminating engine oil with fuel. I will say that oil analysis hasn't shown it to be a major concern but it is present and certainly can't help lubrication or protection. It's more of a benefit of you do a lot of idling or mainly city driving. Since I've installed the tune and delete my wife has averaged a solid 29-31 mpg with almost exclusively city diving. Where before she was getting 24 or so. We haven't had the chance to perform a proper highway evaluation but I'm looking forward to it.
